The model is based on a Perkin Elmer OURA2-SP2, a homebuilt 4-photon microscope that we have previously constructed. It includes a pair of 690nm laser diode bars for excitation, two PMTs for detection, and a shutter for synchronizing with the laser excitation. This new model adds a 32-bit Arduino board, and provides a more efficient arrangement for the laser excitation circuit and the PMT electronics. Additionally, an 8-bit resolution microcontrollers are used for calibration and PMT control.
The requirements for optical alignment are relatively simple; the detector viewport should be centered and tilted to align with the zero-degree pointing direction of the microscope’s laser beams. This is actually relatively straightforward to achieve. The remaining optical alignment requirements are more difficult to account for; the x-y stage is first oriented so that the detector viewport is positioned over the sample plane. Next, the z stage is levelled using the digital micrometer. It should be noted that two-photon microscopy requires beam alignment under fluoranthene cy5 to generate two-photon fluorescence.
Some additional features and documentation are included, including a complete and detailed instruction manual, a bill of materials, schematics for the major components, and an extremely detailed specification and drawing of the complete system that is included with the model.
